Are you using a default aircraft or an add-on?

Try resetting all controls to default.  Then, when you are on the runway, press '.' (the period) to try and release the brakes.

Thanks your kind reply. I just discovered I had another contoller (for model sim planes) connected to another USB port and this caused the conflict. So all is fine now and I can take off in a jiffy without brakes. Anyhow I appreciate very much your kind efforts to solve my problem.
