Adjust the throttle to about 60-70% then trim the aircraft into a neutral flying position so you can take you hand off the stick and she carries on straight and level

Okay do you know how to trim the aircraft correctly? The most tricky bit is vertical, use 1 and 7 on the numeric pad to adjust the elevator trim to smooth out the flight, it won't be instant and takes a little practice but can be easily mastered.
Once you've got the climb/descent sorted against your throttle settings the rest is easy.
I assume you're using the default Jumbo?

There is normally a trim wheel on the 2d panel. Silly question but do you have the Numeric Pad activated (little light on on your keyboard?), if so turn it off, for some reason FS is funny about this.....

Have you made sure that numlock is off?
You could also use AP to cruise like the pros, but that's no fun of course
